反相高效液相色谱法测定人血清中替考拉宁的浓度及其应用 被引量:14

Determination of teicoplanin in human serum by RP-HPLC and its application

摘要 目的:建立测定复杂用药环境下人血清中替考拉宁浓度的反相高效液相色谱法。方法:色谱柱为Agilent ZORBAX SB-C_(18)柱(250mm×4.6mm,5μm),流动相为乙腈-0.02mol·L^(-1)的NaH_2PO_4溶液(24∶76,H_3PO_4调pH值为2.02),流速1.0mL·min^(-1),检测波长240nm,柱温28℃。结果:替考拉宁在2.01~80.67μg·mL^(-1)(r=0.998 9,n=7)范围内浓度与校正因子f(替考拉宁峰面积/哌拉西林的峰面积)成良好的线性关系,定量限为50.5ng,替考拉宁平均提取回收率为87.17%,方法平均回收率为100.7%,(RSD=1.93%,n=9)。结论:该法操作简便、灵敏、准确,适用于替考拉宁血药浓度的测定。 OBJECTIVE To establish a RP-HPLC method for determination of teicoplanin in serum in complicated condi- tions. METHODS An Agilent ZORBAX SB-Cls column was used as the chromatographic column, the mobile phase consisted of a mixture of acetonitrile-0. 02 mol. L-1 Nail2 PO4 (24:76, pH was adjusted to 2. 02 with H3 PO4 ). The flow rate was 1 mL. min-1 , the detection wavelength 240 nm, and column temperature 28 ℃. RESULTS There was a good linear relationship be- tween the concentration of teicoplanin and correction factor (f, peak area value of teicoplanin/peak area value of piperacillin) within the range of 2. 01 - 80. 67 g· mL- 1 (r = 0. 998 9, n = 7), limit of quantitation was 50. 5 ng. The average extraction recov- ery was 87. 17 %, and the average method recovery was 100. 7 % (RSD = 1.930%, n = 9). CONCLUSION The method is sim- ple, sensitive and accurate, which can be used for the assay of teicoplanin in serum under complicated conditions.
