摘要
iTRAQ技术即同位素相对标记与绝对定量技术,主要应用于蛋白质的鉴定与定量分析,结合串联质谱分析,已成为目前蛋白质组学研究的主要手段之一。因寄生虫在不同发育阶段表达虫体蛋白的差异和独特的生活史,其对宿主的选择性、致病性具有较大差异,将iTRAQ技术运用于寄生虫蛋白质组学研究,能有效找出不同阶段寄生虫表达的差异蛋白及感染后宿主表达的差异蛋白,并能对其进行高精度定量分析,这对进一步探索寄生虫生活史、研究与宿主的互作机制提供了技术支撑,同时对寄生虫病的快速诊断及治疗,寻找新的药物靶点等具有重要意义。文章简述了iTRAQ定量蛋白质组学技术及其在寄生虫病研究方面的应用进展。
iTRAQ means isobaric tags for relative and absolute quantitation, which become one of the most important techniques of proteomics research when combined with tanden mass spectrometry. Parasite exhibits the difference on selectivity and pathogenicity of host due to the different stages of protein expression and the unique life history of parasite. The iTRAQ can effectively identified the protein expression or secretion between different stages, and the resulting data could be used for high precision quantitative analysis. All of the results are the key factors to explore the infection mechanism of parasites, which is significant for our further study of its life history. The iTRAQ will facilitated development of the diagnosis and treatment of parasitic disease, and may help us to find new drug targets or vaccine candidates. This paper briefly describes the research progress on iTRAQ quantitative proteomics technology and its achievements on parasitic disease.
