甘蓝DH株自交后代2个变异株群差异性分析 被引量:4

Difference Analysis on Two Variant Group from Self-crossed Progeny of DH Plant in Cabbage

摘要 以甘蓝F1游离小孢子1个胚状体再生植株自交后代分离出DH12-8-Z型和DH12-8-T型2个变异株群为材料,分析2个株群在幼苗期、莲座期、结球期的植株形态,叶片组织结构,光合色素质量浓度和生理生化指标的差异性。结果显示:(1)形态特征上DH12-8-T型植株比DH12-8-Z型植株幼苗矮小、叶片多、叶柄短缩、节间距小、叶片小而厚、边缘翻卷呈"托盘型";结球植株矮小、外叶簇生严重、叶片褶皱、叶中脉变粗、叶色灰绿、叶球小且紧实。(2)相比DH12-8-T型植株幼苗叶片叶绿素a、叶绿素b、总叶绿素和类胡萝卜素质量浓度较低;幼苗叶片组织表皮细胞较小且排列整齐紧凑,海绵组织多呈椭圆或近椭圆形,细胞明显较大,叶脉中维管束组织发达,特别是木质部导管明显粗且多。(3)DH12-8-T型植株比DH12-8-Z型植株可溶性糖和可溶性蛋白质质量分数及POD和CAT活性均较高;莲座期SOD活性较低,莲座期和结球期MDA质量摩尔浓度较低。研究表明,DH12-8-Z型植株和DH12-8-T型植株在形态和生理生化等方面都存在显著差异,两者可能是基因型不同的植株群体。 Two variant plants DH12-8-Z and DH12-8-T were taken as materials,which were segregated from self-crossed progeny of plant regeneration of one embryoid through isolated microspore culture of F1.The deferences of plant morphology,leaf tissue structure,photosynthetic pigments mass concentration,differences of physiological and biochemical indexes between DH12-8-Z type and DH12-8-T-type were measured and analysed during the stages of seedling,rosette and heading.The results showed that:(1)In morphological characteristics,DH12-8-T-type showed shorter seedlings,smaller section space,variate leaves(contain more,smaller and thicker leaves,leaves rolled as"tray-type",etc.)compared with DH12-8-Z-type.And at heading stage,DH12-8-T-type had more outer leaves,leaves folded more seriously,thicker leaf veins,gray-green leaves,more head weight and matured earlier than those of DH12-8-Z-type.(2)Compared with DH12-8-Z-type,seedling of DH12-8-T-type had less mass concentration of chlorophyll a,chlorophyll b,the total mass concentration of chlorophyll and carotenoid;the organizational structure of DH12-8-T-type leaves showed that epidermal cells were smaller,neat and compact,spongy tissue s were mostly oval or nearly oval,cells were larger,vasculartissue of leave veins more developed,particularly xylem conduits were more and thicker.(3)The soluble sugar and soluble protein mass fraction,POD,CAT activity of DH12-8-T-type plants in each developmental period were higher than those of DH12-8-Z-type;the SOD activity of DH12-8-T-type plants was lower that of DH12-8-Z-type plants at the rosette stage;the MDA mass molality of DH12-8-T plants was higher that of DH12-8-Z-type plants at seedling stage,on the contrary,the MDA mass molality of DH12-8-T plants at rosette and heading stage were lower.In conclusion,there are significant differences in the plant form growth,physiology and biochemistry between DH12-8-Z type and DH12-8-T type during the growth period,the two variant group may be variability groups of different genotypes.
