摘要
目的研究黄芩汤对葡聚糖硫酸钠(DSS)诱导的结肠炎小鼠模型CD98的干预作用。方法 32只C57BL/6小鼠随机分为对照组、DSS组、DSS+黄芩汤组、DSS+美沙拉嗪组,每组8只。用3.5%DSS制成结肠炎小鼠模型后,分别-1-1-1-1予以黄芩汤(130 mg·kg·d)和美沙拉嗪(7.4 mg·kg·d)治疗7 d。检测结肠长度和结肠组织髓过氧化物酶(MPO)活性,免疫组化和Western blot法检测小鼠结肠上皮CD98表达。结果 CD98在结肠炎小鼠结肠上皮中表达增高,黄芩汤增加结肠炎小鼠结肠长度,降低MPO活性,抑制CD98过表达。结论黄芩汤可抑制DSS诱导的结肠炎小鼠结肠上皮高表达CD98,这可能是黄芩汤治疗炎症性肠病的机制之一。
Objective To evaluate the intervention of Huangqin decoction on CD98 expression in mouse colitis induced by dextran sodium sulfate(DSS). Methods Thirty-two C57BL/6 mice were randomly divided into control, DSS, DSS + Huangqin decoction and DSS + mesalazine groups. After the colitis models were established by 3.5% DSS, the latter two groups were-1-1-1-1treated with Huangqin decoction(130 mg·kg-1 ·d-1) and mesalazine(7.4 mg·kg-1 ·d-1), respectively. The colon length and myeloperoxidase(MPO) activity in colon tissues were measured, and CD98 expression of colonic epithelia was detected by immunohistochemistry and Western blot. Results CD98 overexpression of colonic epithelia was observed in colitis mice. Huangqin decoction increased the colon length, attenuated the MPO activity, and inhibited the CD98 overexpression in colitis mice. Conclusion Huangqin decoction can inhibit CD98 overexpression of colonic epithelia in murine colitis, which might be one of the therapeutic mechanisms for inflammatory bowel diseases.
