灯盏细辛活性部位对神经细胞α3尼古丁受体蛋白水平的影响 被引量:1

Influence of the Active Parts Extracted from Herba Erigerontis on the Protein Level of α3 Nicitinic Receptor Subunit in Human Neuroblastoma SH-SY5Y Cells

摘要 目的研究灯盏细辛醇提取乙酸乙酯萃取物不同活性部位对神经母细胞瘤细胞(SH-SY5Y)中尼古丁乙酰胆碱能受体(Nicotinic acetylcholine receptor,nAChR)蛋白水平的影响及其对抗β-淀粉样蛋白(Aβ)细胞毒性的作用机制,以探讨其在阿尔茨海默氏病(Alzheimer's disease,AD)治疗中的作用。方法选用一定浓度的灯盏细辛不同部位及Aβ25-35处理SH-SY5Y细胞,用比色法测定培养细胞MTT还原率,蛋白质印迹法测定尼古丁受体α3亚单位蛋白水平。结果灯盏细辛醇提取乙酸乙酯萃取物不同部位中,经凝胶柱分离的第一区段、该区段经反相硅胶柱分离的低极性部位及乙酸乙酯萃取后再以正丁醇获得的萃取物,经凝胶柱分离的第四区段经反相硅胶柱分离出的低极性部位能上调SH-SY5Y细胞中α3nAChR蛋白水平;同时能减弱Aβ25-35的细胞毒性作用。结论灯盏细辛醇提取乙酸乙酯萃取物分离部位可能是中药灯盏细辛的重要的活性部位,在阿尔茨海默氏病的治疗中可能起到一定作用。 Objective To investigate the effect of the active parts extracted from Herba Erigerontis on the protein level of α3 nicitinic receptor subunit in human neuroblastoma(SH-SY5Y) cells and against the neurotoxicity in SH-SY5Y cells induced by beta-amyloid peptide(A).Methods The SH-SY5Y cells were treated by a certain safe concentration of the active parts extracted from Herba Erigeronitis,and exposed to Aβ25-35.MTT reduction was detected by photomatric assay,and the protein level of α3 nAChR subunit was detected by Western-blotting.Results The active parts extracted from Herba Erigerontis stimulated upregulation of α3 nAChR protein in the cultured cells and prevented the MTT reduction resulted from Aβ.Conclusion The active parts extracted from Herba Erigerontis may prevent neurotoxicity of Aβ by the upregulation of nAChR,which might play an important role in therapeutic strategy of AD.
