摘要
以T3菌株为代表菌株对中华鳖嗜水气单胞菌的生物学特性进行初步研究。对中华鳖嗜水气单胞菌的培养特性比较发现:本菌在鲜血琼脂和TSA固体培养基上生长较好,在TSB液体培养基和BHI液体培养基中生长较好;测定中华鳖嗜水气单胞菌在TSB液体培养基中的生长曲线表明:培养16~20h活菌数达到最高峰,24h后活菌数大幅下降,这一生长规律为中华鳖嗜水气单胞菌的大规模培养奠定了基础;中华鳖嗜水气单胞菌在生理盐水、磷酸盐缓冲液和TSB液体培基中4℃保存的存活曲线表明:本菌在以上介质中前期不易死亡,后期死亡较快。并利用分光光度计测定法解决了该菌即时计数的问题,为中华鳖嗜水气单胞菌的动物实验的开展奠定了基础,本研究测定了该菌对中华幼鳖和银鲫的致病性,其LD50分别为8.40×108CFU和8.63×108CFU。
T3 Aeromonas hydrophila was used as model strain to study the biological features of Aeromonas hydrophila.The growth of the strain on the BA and TSA solid medium is better than other solid medium,and the growths in TSB and BHI broth are better than other fluid medium.The growth curve indicates that the number of viable T3 Aeromonas hydrophila growing in the TSB broth reaches the peak after cultured 16~20 hours and the viable strain decreases rapidly after cultured 24 hours.The discovery of Aeromonas hydrophila growth character establishes the foundation of large-scale culture of the bacteria.Survival curve indicates that the strain preserved in physiologic saline,PBS and TSB broth at 4℃ can survive well at protophase but difficult survive at anaphase.Moreover spectro photometer assay of the strain solves the problem of counting of the viable strains which settles the foundation of animal experiment of Aeromonas hydrophila.The pathogenicity to china immature soft-shell turtle and johnny carp of the T3 Aeromonas hydrophila is researched and LD50 of T3 Aeromonas hydrophila to china immature soft-shell turtle and johnny carp are 8.40×108 CFU and 8.63×108 CFU respectively.
