摘要
目的研究小刺猴头菌醇提物(EHCP)对阿尔茨海默病(AD)模型小鼠的治疗作用及作用机制。方法给小鼠连续15d腹腔注射D-半乳糖120mg·kg-1和亚硝酸钠90mg·kg-1,以制备AD小鼠模型,在造模7d后,连续45d给实验组小鼠按1g.kg-1灌胃10%的EHCP。通过Morris水迷宫、酶学检测及NGF免疫组化染色,观察EHCP对AD模型小鼠学习记忆、脑内乙酰胆碱酯酶(AchE)活性和神经生长因子(NGF)含量的影响。结果D-半乳糖和亚硝酸钠联合诱发的AD模型鼠寻找潜伏期明显延长、AchE活性及NGF蛋白表达水平明显下降;EHCP可使AD模型鼠的寻找潜伏期分别由第3d的(48.7±35.5)s和第4d的(49.5±32.1)s缩短至(18.5±7.2)s和(15.4±9.2)s(P<0.05);酶学检测结果表明EHCP可升高AD模型鼠脑内AchE活性(P<0.05);免疫组化结果显示EHCP可增加其脑内NGF蛋白表达水平(P<0.01)。结论EHCP可有效的提高AD小鼠的判断及记忆能力,其机制可能与EHCP保护神经细胞及促进神经细胞再生等相关。
Objective To study the effects and mechanism of ethanol extract from Herieium captmedusae (Bull. ex Fr.) Pers on the Alzheimer's disease (AD) model mice. Methods The mice were treated with D-galactose (120 mg·kg^-1·d^-1,ip,15 d) and NaNO2 (90 mg·kg^-1·d^-1 1 ,ip,15 d) to make AD model mice. The test group mice were treated with 10% ethanol extract from Herieium captmedusae (Bull. ex Fr.) Pets (1 g·kg^-1·d^-1,po,45 d) since d8 when made model. The Morris water-maze test,AchE activity test and the immunohistochemical analysis of NGF in the brain were carried out to evaluate the effects of ethanol extract from Herieium captmedusae (Bull. ex Fr, ) Pers on AD model mice. Results Escape latency time was prolonged,AchE activity and level of NGF were reduced in AD model mice induced with D-galactose and NaNO2. Ethanol extract from Herieium eaptmedusae (Bull. ex Ft. ) Pers significantly reduced the escape latency time from (48.7±35.5) s to (18.5±7.2) s on the third day and from (49.5±32.1) s to (15.4±9.2) s on the fourth day (P〈 0.05),in creased AchE activity by enzymology test (P 〈 0.05) and level of NGF by immunohistochemieal analysis (P 〈 0.01 ) in the brain of AD model mice. Conclusion Ethanol extract from Herieium eaptmedusae (Bull. ex Fr.) Pers can increase judgement and memory of AD model mice. And the mechanism is probably relevant to protect nerve cell and promote nerve cell regeneration.
