Effect of Temperature on the Growth Dynamics of the Nuclear Polyhedrosis Virus from Ectropis obliqua hypulina Wehrli in its Host Larvae

摘要 多角体计数、对流免疫电泳、单向免疫扩散及火箭免疫电泳测定的结果表明:26℃适于茶尺蠖核型多角体病毒(EoNPV)的增殖。多角体含量或其相对值随着时间的推移而增长,并渐趋于平稳,两者间呈Logistic曲线关系。单位体重或单头幼虫所含的多角体数量(y1或y2)、扩散环直径(y3)和火箭峰值(y4)与时间(t)的关系式分别为:y1=(8.1481)/(1+EXP(9.4210-0.0608t))×109PIB/克;y2=(6.1596)/(1+EXP(5.4809-0.0376t))×108PIB/头,y3=(1.4)/(1+EXP(2.710-0.015t))cm;y4=(3.52)/(1+EXP(4.580-0.040t))cm。但30℃下,EoNPV增殖严重受抑制,饲毒后24~168小时内难以测出多角体,其后多角体含量也极显著低于26℃,乃至难以被三种免疫测定法测出。 Effect of temperature on the growth dynamics of the nuclear polyhedrosis virus form Ectropis obliqua hypulina (EoNPV) in its host larvae was studied through polyhedra counting, counter immuno-electrophoresis, single immunodiffusion and rocket immuno-electrophoresis tests. The propagation of the virus in the host larvae fed with the NPV at the concentration of 2.6 10~7PIB/ml and reared at 26℃ was normal. The polyhedra titer or its relative equivalences increased positively with the time after inoculation and then gradually reached a plateau. The growth dynamic curve obeyed Logistic curve. The regression equations of the relationship between the number of the polyhedra per gram larval weight (y_t), the number of the polyhedra per larva (y_2), immunodiffusion ring diameter (y_3) and rocket highness (y_) and the time after inoculation(t)were obtained as follows: y_=8.1481/(1+EXP(9.4210-0.0608t))×10~9PIB/g; Y_2=6.1596/(1+EXP(5.4809-0.0376t)×10~8PIB/Jarva; y_3=1.4/(1+EXP(2.710-0.015t) cm; y_4=3.52/(1+EXP(4.580-0.040t))Cm. When the host larvae were fed with the virux and reared at 30℃, the propagation of EoNPV was obviously inhibited. When the host larvae were exposed to the virus from 24 to 168 hr, no polyhedra could be detected. Afterwards the concentrations of the polyhedra were also significantly lower than that reared at 26℃, and the polyhedra could not be detected by three immunoassay methods.
