CpG基序在人食管癌Eca-109细胞疫苗中的佐剂效应

摘要 目的:探讨CpG基序在人食管癌Eca-109细胞疫苗中的佐剂效应。方法:培养Eca-109细胞,用酸洗法收集细胞膜表面小肽,再用去盐、浓缩、超滤法纯化这些小肽后,以此刺激人外周血树突状细胞(DC)作为人食管癌疫苗,不用小肽刺激的DC作为对照疫苗,并用这些疫苗加佐剂CpG基序激活纯化的T细胞。实验分组:①CpG基序加食管癌疫苗组(CpG-Ve);②CpG基序加食管癌对照疫苗组(CpG-Vc);③单纯食管癌疫苗组(Ve);④单纯食管癌对照疫苗组(Vc);⑤单纯CpG基序刺激组;⑥直接用纯化的T细胞加Eca-109细胞作效应对照组。用3H渗入法对各组行T细胞增殖试验,用51Cr释放试验测定各组细胞毒性T淋巴细胞(CTL)的活性。结果:CpG-Ve组的T细胞增殖活性和CTL对Eca-109细胞杀伤率均高于其余各组(P<0.01)。结论:CpG基序可能是人食管癌疫苗研究中的有效佐剂。 Aim: To study the adjuvant effects of CpG motif in vitro on human esophageal cancer Eca-109 cell vaccine. Methods: Eca-109 cells were cultured and the peptides on cell membrane were collected by mild acid wash method. The obtained peptides were purified by removing salt, concentrating and ultrafiltrating. Matured DC activated by the peptides were used as human esophageal tumor vaccine. The purified T cells in vitro were activated by the above tumor vaccine combined with adjuvant of CpG. T-cell proliferation was analyzed by determining 3H thymidine incorporation and CTL response was assa)ed by ^51 Cr-release. Results: CpG markedly enhanced T-cell proliferation and CTL response( P 〈 0.01 ). Conclusion : CpG may be the optimal adjuvant of human esophageal cancer vaccine.
