血塞通注射液对梗阻性黄疸肾损伤的保护机制 被引量:1

摘要 目的 :观察血塞通注射液对梗阻性黄疸引起的肾损伤的保护作用 .方法 :健康日本大耳白兔 4 8只 ,随机分为 2组 :A组为胆总管结扎组 2 4只 (obstructivejaundice,OJ) ;B组为胆总管结扎 +血塞通注射液组 2 4只 .每组又进一步分为 4个亚组 .建立胆道梗阻动物模型后 ,在术后不同时期取材测定血清总胆红素 (T BIL)、血肌苷 (Cr)和尿素氮 (BUN)值 ;用免疫组化法检测肾组织Bax、Bcl 2的表达情况 .结果 :胆道梗阻后A、B两组的肾脏均有Bcl 2和Bax蛋白表达 ,Bax蛋白在梗阻 9d时达高峰 .Bcl 2蛋白在梗阻 2 1d达高峰 .凋亡指数(AI)随Bax的升高而升高 ,随Bcl 2的升高而降低 .B组Bax蛋白表达较同时相组弱 ,而Bcl 2蛋白表达较同时相组强 .B组AI值较A组降低 .结论 :血塞通注射液对梗阻性黄疸肾损伤有保护作用 ,其机制可能是通过上调肾组织Bcl 2蛋白表达 。 AIM: To study the protective effects of XueSaiTong Injection on the renal injury caused by obstructive jaundice (OJ) in rabbits. METHODS: Forty eight rabbits were first randomly divided into two groups: the group of OJ (group A, 24 rabbits) and the group of OJ treated with XueSaiTong Injection (group B, 24 rabbits). Then each group was further subdivided into four subgroups and all the rabbits used in this study were raised under the same condition. Auto biochemical analysis equipment was used to examine the T BIL, Cr and BUN in serum at different times and immunohistochemistry was used to determine the expressions of Bax and Bcl 2 on renal. RESULTS: Expressions of Bcl 2 and Bax on the renal tissues were observed following OJ. The expression of Bax reached the peak value at the 9th d and the highest expression of Bcl 2 was observed at the 21st d. The apoptosis index (AI) increased with the increase of Bax expression but decreased with Bcl 2 strong expression. A lower expression of Bax and a higher expression of Bcl 2 were found in group B compared with those of group A. The AI value was also lower in group B than that in group A. CONCLUSION: XueSaiTong Injection has some protective effects on the renal injury caused by OJ, for XueSaiTong Injection may up regulate the expression of Bcl 2 and down regulate the expression of Bax in renal tissues.
