摘要
目的:建立RP-HPLC同时测定壮药清解雾化液中绿原酸、 芦丁、 金丝桃苷、 异槲皮苷4种指标成分的含量测定方法.方法:应用日本岛津LC-20A高效液相色谱仪,SPD-20A检测器,WondaCract ODS-2(4.6 mm×250 mm,5μm)色谱柱,以乙腈溶液(A)-0.2%磷酸水溶液(B)为流动相,梯度洗脱;流速1.0 mL/min,检测波长355 nm,柱温35℃.结果:绿原酸、 芦丁、 金丝桃苷、 异槲皮苷的进样量分别在2.22~5.55μg、0.330~0.825μg、0.208~0.52μg、0.3485~0.8713μg范围内线性关系良好(R2≥0.9997);加样回收率为97.5% ~102.35%,RSD值为1.15% ~2.14%.结论:该方法专属性、 稳定性、 重复性高,灵敏、 简便,可应用于壮药清解雾化液制备工艺质量控制.
Objective To develop an RP-HPLC method for simultaneous determination of chlorogenic acid,rutin,hyperoside and isoquercitrin in Qingjie Atomized Liquid.Methods Separation was carried out an WondaCract ODS-2(4.6 mm×250 mm,5μm),eluted with mobile phase of 0.2%phosphoric acid solution(B)-acetonitrile(A)in gradient mode.The flow rate was 1.0 mL/min.Column temperature was 35℃and detection wavelength was set at 355 nm.Result Linearity was good for chlorogenic acid,rutin,hyperoside and isoquercitrin in range of 2.22~5.55μg,0.330~0.825μg,0.208~0.52μg,0.3485~0.8713μg.respectively,Average recoveries of 4 compounds were 97.5%-102.35%,RSD(1.15%-2.14%).Conclusion The method is accurate,stable,repeatable,sensitive and simple,can be used for quality control of Qingjie Atomized Liquid.
