摘要
目的:通过对比忍冬枝枯病感病植株和健康植株4个花期金银花中绿原酸含量与绿原酸合成相关酶编码基因转录水平,初步探究了忍冬枝枯病影响金银花品质的分子机制。方法:以5年生忍冬枝枯病感病植株和健康植株头茬花4个花期花蕾为材料,采用高效液相色谱法(high performance liquid chromatography,HPLC)比较绿原酸含量差异;从本实验室的忍冬-枝枯病互作的转录组数据中挑选出与绿原酸合成相关的差异表达基因LjHQT和LjC4H2,通过real-time RT-PCR技术分析了枝枯病对2个基因转录水平的影响。结果:感病植株4个花期金银花中绿原酸的含量均高于健康植株对应花期。对比健康植株、感病植株4个花期中LjHQT基因转录水平下调表达;LjC4H2基因转录水平则为上调表达,其中在青花期,感病植株中LjC4H2基因的相对表达量是健康植株青花期的36.50倍。结论:忍冬枝枯病可以通过影响苯丙素生物合成途径中桂皮酸途径相关酶基因LjHQT和LjC4H2的转录水平来影响金银花中绿原酸的合成。
Objective:To explore the molecular mechanism underline the effect of branch blight on the quality of honeysuckle,we conducted the comprehensive analysis of the content of chlorogenic acid and the transcriptional levels of related genes in the four flowering phases from the disease and healthy plants of L.japonica.Methods:The content of chlorogenic acid in disease and healthy plant materials in the four flowering phases were compared by high performance liquid chromatography(HPLC).The chlorogenic acid synthetic related differentially expressed genes were selected from the transcriptome data of the interaction between the pathogen caused branch blight disease and its host L.japonica,and the transcriptional levels of genes LjHQT and LjC4H2 were quantified by real time RT-PCR.Results:The content of chlorogenic acid in the four flowering phases of disease plants were higher than that in the healthy plants.The transcriptional level of LjHQT gene in the four flowering phases of disease plants was down-regulated compared with that of healthy plants.However,the transcriptional level of LjC4H2 gene was up-regulated.The transcriptional level of LjC4H2 gene in the four flowering phases of disease plants was higher than that of healthy plants,especially in the green flower period.The transcriptional level of LjC4H2 gene in disease plants was 36.50 times higher than that of healthy plants.Conclusion:Branch blight disease can influence the synthesis of chlorogenic acid in L.japonica by affecting the transcriptional levels of cinnamic acid pathway related synthetase genes LjHQT and LjC4H2 in the phenylpropanum biosynthesis pathway.
