摘要
目的建立易黄汤物质基准的最佳制备工艺。方法采用单因素试验法和正交试验法,以小檗碱、没食子酸、毛蕊花糖苷和固含物的转移率为评价指标,采用HPLC法测定含量,对易黄汤前处理、煎煮、过滤、浓缩和干燥等工序的参数进行优选。结果易黄汤物质基准最优的制备工艺为:取炒白果粉碎过1号筛,酒车前子包煎,同其它饮片置于药罐中煎煮2次,第1次加8倍量水,浸泡30 min,加盖煎煮30 min;第2次取药渣再加7倍量水煎煮25 min,趁热滤过。合并两次滤液减压浓缩至1.0 g/mL饮片,冷冻干燥48 h,得冻干粉约8 g,置于20 mL棕色玻璃西林瓶中密封保存,即得易黄汤物质基准。结论优选的易黄汤物质基准制备工艺稳定可行。
Objective To establish the best preparation process of Yihuang Decoction(易黄汤)material standard.Methods Single factor test method and orthogonal test method were used to evaluate the transfer rate of berberine,gallic acid,verbascoside and solid content.The content was determined by HPLC method.The pretreatment,decoction and filtration of Yihuang Decoction,concentration and drying process parameters were optimized.Results The optimal preparation process of Yihuang Decoction material standard were:take the fried ginkgo and crush it through the No.1 sieve,and fry the psyllium psyllium for frying,then place it in a medicine pot with other decoction pieces for two times,add 8 times the amount of water for the first time,soak for 30 minutes,cover and decoct for 30 minutes,take the medicine residue for the second time and add 7 times the amount of water to decoct for 25 minutes.The two filtrates were combined under reduced pressure and concentrated to 1.0 g/mL,and freeze-dried for 48 hours to obtain about 8 g of freezedried powder,which was placed in a 20 mL brown glass vial and seal and store to obtain the substance standard of Yihuang Decoction.Conclusion The optimal preparation process of Yihuang Decoction is stable and feasible.
