喜炎平注射液对重症手足口病幼鼠模型血浆代谢组学的影响

Effect of Xiyanping Injection on plasma metabolomics in young rats with severe hand-foot-and-mouth disease

摘要 目的探讨喜炎平注射液对重症手足口病幼鼠模型血浆代谢组学的影响。方法将45只1周龄SD大鼠按随机数字表法分为对照组(control,15只)、模型组(model,15只)和喜炎平组(XYP,15只),对照组大鼠不做任何处理,模型组大鼠腹腔注射肠道病毒EV71-BJ制备手足口病幼鼠模型,喜炎平组大鼠在腹腔注射EV71-BJ基础上,尾静脉注射100mg/kg的喜炎平。模型制备1天后采集血浆标本,利用超高压液相色谱串联四级杆飞行时间质谱技术(UPLC-Q-TOF-MS/MS)测定血浆代谢组学,利用Metaboscape 3.0软件比对鉴定内源性代谢物。通过MetaboAnalyst 5.0网站进行多元回归分析,包括主成分分析(principal components analysis,PCA)和偏最小二乘法判别分析(partial least squares discriminant analysis,PLS-DA)模式识别各代谢物的变化,筛选相关差异代谢物[P<0.05、变量权重值(variable important in projection,VIP)>1.5],并进一步通过KEGG分析相关代谢通路。结果PCA和PLS-DA模式识别结果显示,三组之间表现出聚类型分布,PLS-DA模式识别优于PCA;三组之间检出64个差异代谢物,其相关代谢途径包括精氨酸生物合成、精氨酸和脯氨酸代谢、丙氨酸、天冬氨酸和谷氨酸代谢和鞘脂代谢。结论喜炎平注射液影响重症手足口病幼鼠模型血浆氨基酸代谢和鞘脂代谢过程。 Objective To investigate the effect of Xiyanping injection on plasma metabolomics in in young rats with severe hand-foot-and-mouth disease.Methods Forty-five one-week-old SD rats were randomly divided into control group(control,15 rats),model group(model,15 rats)and Xiyanping group(XYP,15 rats).Rats in the model group were injected intraperitoneally with enterovirus EV71-BJ to prepare models.Rats in the XYP group were injected with 100 mg/kg of Xiyanping via the tail vein,beside EV71-BJ intraperitoneally injection.Plasma samples were collected 1 day after model preparation and metabolomics was determined for metabolomics by UPLC-Q-TOF-MS/MS.The endogenous metabolites were identified by Metaboscape 3.0 software database.Multiple regression analysis was carried out through MetaboAnalyst 5.0,including principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)to identify the changes of metabolites,screen the related differential metabolites(P<0.05 and VIP>1.5),and then further analyze the related metabolic pathways through KEGG.Results The PCA and PLS-DA pattern recognition showed that there was a cluster type distribution between the three groups.64 differential metabolites were respectively detected between the three groups,and their related metabolic pathways included arginine Biosynthesis,arginine and proline metabolism,alanine,aspartate and glutamate metabolism and sphingolipid metabolism.Conclusions Xiyanping injection affects the metabolism of amino acids and sphingolipids in plasma in young rats with severe hand-foot-and-mouth disease.
