摘要
目的:采用网络药理学方法,探讨三金排石加减方治疗石淋的作用机制。方法:利用TCMSP、HERB以及SwissTarget数据库筛选核心处方中药物的活性成分与作用靶点,并从CTD、GeneCards、OMIM、TTD数据库中获取石淋的疾病靶点,进一步构建药物-靶点-疾病关系网络,运用String数据库构建PPI网络,通过Metascape平台对交集靶点进行GO功能、KEGG通路富集分析,利用Autodock vina软件进行分子对接模拟验证,得出结果。结果:处方的分析,去除重复值后,共有113个活性成分、1470个靶点,疾病靶点365个,药物与疾病交集靶点101个,得出作用于疾病的化合物成分有55个。结论:通过网络药理学研究发现,三金排石加减方治疗石淋的作用机制可能是其有效成分,通过抑制炎性反应、氧化应激反应等靶点,调控NF-KB信号通路、P38MAPK通路、Th17细胞分化等实现的。
Objective:Using Professor Mi's San Jin Pai Shi Addition and subtraction prescription for the treatment of Urolithiasis,using network pharmacology methods to explore its mechanism of action in treating Urolithiasis.Research Methods:Using the TCMSP,HERB,and the SwissTarget database to screen the active ingredients and action targets of the drugs in the core prescriptions,And obtained the disease targets of gonorrhea from the CTD,GeneCards,OMIM,TTD databases,Further,the construction of the drug-target-disease relationship network,Build a PPI network using the String database,GO function,KEGG pathway enrichment analysis of intersection targets by Metascape platform,Molecular docking simulation validation using Autodock vina software,Get the result.Results:After the analysis of the prescription of repeat value,there were 113 compounds,1,470 targets,365 disease targets,101 drug and disease intersection targets,and 55 compound components acting on the disease.Conclusion:Through network pharmacology research,the mechanism of action of San Jin Pai Shi Addition and subtraction prescription in treating Urolithiasis may be its effective component,which regulates the NF-KB signaling pathway,P38MAPK pathway,Th17 cell differentiation,and other targets by inhibiting inflammatory and oxidative stress responses.
