基于UPLC-Q-TOF-MS的网果酸模化学成分分析 被引量:2

摘要 目的:分析网果酸模Rumex chalepensis Mill.的主要化学成分。方法:采用UPLC-Q-TOF-MS分析方法,色谱柱为Agilent ZORBAX Extend-C_(18)柱(250 mm×4.6 mm,5μm),流动相为水(含10 mmol/L醋酸铵)-乙腈,梯度洗脱,流速1.0 mL/min,柱温30℃,进样量5μL,电喷雾离子源(ESI),干燥器温度(Gas Temp)350℃,干燥气流量10 L/min,雾化器压力45 psi,毛细管电压4500 V,扫描范围m/z 100~1500,正、负离子模式下对色谱流出物进行质谱检测,利用对照品、二级质谱信息及文献对各主要色谱峰进行归属、鉴定。结果:共鉴定化合物18个,其中结合蒽醌化合物3个,游离蒽醌化合物3个,萘类化合物4个,黄酮类化合物7个,木脂素类1个,首次鉴定化合物12个。结论:采用UPLC-Q-TOF-MS可快速分析网果酸模主要化学成分,为网果酸模质量控制及申报地方标准奠定基础。 Objective:To analyze the chemical constituents of Rumex chalepensis.Methods:Using UPLC-Q-TOF-MS analysis method,chromatographic column was Agilent ZORBAX Extend-C_(18)(250 mm×4.6 mm,5μm),the mobile phase was water(containing 10 mmol/L ammonium acetate)-acetonitrile with gradient elution,the flow rate was 1.0 mL/min,the column temperature was 30℃and the injection volume was 5μL.Using electrospray ion source(ESI),dryer temperature(Gas Temp)was 350℃,drying gas flow rate was 10 L/min,atomizer pressure was 45 psi,capillary voltage was 4500 V,scanning range was m/z 100~1500,the chromatographic effluents were detected by mass spectrometry in positive and negative ion mode,the main chromatographic peaks were classified and identified by reference,secondary mass spectrometry and literature.Results:A total of 18 compounds were identified,including 3 combined anthraquinones,3 free anthraquinones,4 naphthoquinones,7 flavonoids and 1 lignans.Twelve compounds were identified for the first time.Conclusion:UPLC-Q-TOF-MS can be used to analyze the main chemical constituents of Rumex chalepensis rapidly,it can lay a foundation for the quality control and declaration of local standards of Rumex chalepensis.
