金银花不同采收期指纹图谱及成分变化情况研究 被引量:13

摘要 目的:建立不同采收茬期的金银花HPLC指纹图谱,并对其化学成分进行动态分析。方法:色谱柱为Symmetry C_(18)(250 mm×4.6 mm,5μm)柱;流动相为乙腈-0.2%甲酸溶液,梯度洗脱;检测波长为245 nm;流速为0.5 mL/min;柱温为38℃。结果:建立了17批不同茬期金银花样品的HPLC指纹图谱,共确定并指认了16个共有峰,相似度为0.910~1.000;主成分分析表明一茬与二茬期采收的金银花样品综合得分空间分布较为接近,可与三茬、四茬样品明显区分。指认的酚酸类、黄酮类、环烯醚萜苷类、咖啡酰奎宁酸类共16个成分的动态变化规律不同。结论:该研究建立了金银花的指纹图谱,探究了酚酸类、黄酮类、环烯醚萜苷类、咖啡酰奎宁酸类等16个成分的动态变化规律,对实现优质金银花规范化生产示范基地建设及其相关成药批次的稳定性研究具有指导意义。 Objective:To establish HPLC fingerprint of Lonicerae Japonicae Flos at different harvesting stages and analyze its chemical constituents dynamically.Methods:The chromatographic column was a Symmetry C_(18)(250 mm×4.6 mm,5μm)column,mobile phase was acetonitrile-0.2%formic acid solution with gradient elution,detection wavelength was 245 nm,flow rate was 0.5 mL/min,column temperature was 38℃.Results:The HPLC fingerprints of 17 batches of Lonicerae Japonicae Flos at different harvesting stages were established,and 16 common peaks were identified.The similarity was ranged from 0.910 to 1.000.The principal component analysis showed that the spatial distribution of the comprehensive scores of the samples collected at the first and second stubble stages was close,and the samples could be clearly distinguished from the third and fourth stubble.The dynamic changes of 16 components such as phenolic acids,flavonoids,iridoid glycosides,caffeylquinic acids were different.Conclusion:In this study,the fingerprint of Lonicerae Japonicae Flos is established,and the dynamic changes of 16 components such as phenolic acids,flavonoids,iridoid glycosides,caffeylquinic acids are explored,which has guiding significance for the construction of standardized production demonstration base of high-quality Lonicerae Japonicae Flos and the stability study of related patent medicine batches.
