川归灵挥发油对重物致软组织损伤大鼠模型的干预作用 被引量:7

Intervention Effect of Chuanguiling Essential Oil on Rat Model with Soft Tissue Injury Induced by Heavy Objects

摘要 目的:探讨川归灵挥发油对重物致软组织损伤大鼠模型的干预作用,明确其潜在作用机制。方法:随机取8只Wistar大鼠为空白组,余下104只采用重物击打法复制非开放性急性软组织损伤模型。翌日,筛选成模大鼠,随机分为模型组、云南白药酊0.1 ml/cm2组,川归灵挥发油0.2 ml/cm2、0.1 ml/cm2组,每组24只。在给药第1 d、4 d和7 d的2 h后每组分别取8只测损伤部位皮温差和肿胀度;眼球取血,测血清IL-1β和bFGF水平;取损伤组织,部分匀浆测PGE2和TNF-α水平,部分HE染色,观察病变情况。结果:与空白组比,给药第1 d、4 d和7 d模型组大鼠皮温差、肿胀程度显著升高,损伤部位组织匀浆PGE2和TNF-α含量显著升高,血清IL-1β含量显著升高,给药第4 d和7 d模型组大鼠血清bFGF含量显著升高。与模型组比,各给药组可不同程度明显降低大鼠第1 d、4 d和7 d皮温差和肿胀度、降低第4 d和7 d匀浆PGE2和TNF-α含量以及血清IL-1β含量、不同程度明显升高大鼠第4 d和7 d血清bFGF的含量。组织病理观察:第1 d,模型组与各给药组大鼠出血、水肿、炎性浸润显著,肌纤维局部断裂、坏死、变形;第4 d,模型组出血、水肿、炎性浸润减轻,仍见坏死。各给药组肌纤维变性坏死、断裂减少,明显改善;第7d,模型组部分肌纤维排列紊乱,较多炎性细胞浸润,散在肉芽组织和瘢痕组织形成。各给药组见正常肌肉组织,炎性细胞极少,纤维排列较整齐。结论:川归灵挥发油对软组织损伤模型大鼠具有改善作用,其改善作用可能与抑制炎症反应(降低皮温差、肿胀度、匀浆PGE2、TNF-α含量以及血清IL-1β含量)和促进血管新生、组织修复(升高血清bFGF含量)有关。 Purpose:To explore the intervention effect of Chuanguiling essential oil on soft tissue injury induced by heavy objects in rat model,and its potential mechanism.Methods:Rats were divided into the control group and the model group,8 in the control group,104 rats were established into non-open soft tissue injury model by heavy objects.The next day,successful model rats were selected and randomly divided into the model group,Yunnan Baiyao group(0.1 ml/cm2)and Chuanguiling essential oil high(0.2 ml/cm2)and low(0.1 ml/cm2)doses of groups,24 rats in each group.The skin temperature difference and swelling degree of the injured area were measured on the 1 st day,4 th day and 7 th day,2 h after administration.Blood samples were taken from eyeballs,IL-1βand bFGF levels in serum were measured.The injured tissues were taken and the levels of PGE2 and TNF-αin homogenate were measured,pathological changes were observed by HE staining.Results:Compared with the control group,the skin temperature difference and swelling degree in the model group were significantly increased,the levels of PGE2 and TNF-αin tissue homogenate of injured sites were significantly increased,the levels of IL-1βin serum were significantly increased on the 1 st,4 th and 7 th day after administration.Compared with the model group,the skin temperature difference and swelling degree were decreased to different degrees on the 1 st,4 th and 7 th day in drug groups,the levels of PGE2 and TNF-αin homogenate on the 4 th and 7 th day were decreased,the level of IL-1βin serum was decreased,and the level of bFGF in serum was increased in different degrees on the 4 th and 7 th day.For histopathological observation,On the 1 st day,hemorrhage,edema,inflammatory infiltration were observed in the model and drugs groups,and the degeneration,necrosis and rupture of muscle fibers were also observed.On the 4 th day,hemorrhage,edema and inflammatory infiltration were alleviated while necrosis was still observed in the model group.The degeneration,necrosis and rupture of muscle fibers in the drugs groups were reduced and obviously improved.On the 7 th day,in the model group,some muscle fibers were disorderly arranged,more inflammatory cells were infiltrated,scattered in granulation tissue and scar tissue.In drugs groups,normal muscle tissue,few inflammatory cells and neat arrangement of fibers were observed.Conclusion:Chuanguiling essential oil can improve the soft tissue injury in model rats.Its improvement may be related to inhibiting inflammation(decreasing skin temperature difference,swelling degree,homogenate PGE2,TNF-αlevel and serum IL-1βlevel),promoting angiogenesis and tissue repair(increasing serum bFGF level).
