摘要
目的:基于硬皮病的中西医临床病证特点,分析现有硬皮病动物模型并为其研究提供思路。方法:通过查阅国内外与硬皮病动物模型相关文献及报道,整理总结硬皮病中西医临床病证表现及诊断标准,并根据中西医临床各项诊断指标,制定相应评价标准。动物模型与西医临床核心指标每符合一项赋值为10%,相关指标每符合一项赋值为4%;与中医临床的主证每符合一项赋值为15%,次证每符合一项为10%,对现有硬皮病动物模型的临床症状吻合度进行评价分析,并且初步提出硬皮病中医病证结合动物模型评价指标。结果:发现现有硬皮病动物模型与临床吻合度较高模型有博来霉素诱导型、UCD200小鸡模型;一般吻合度模型有ROS诱导模型、转基因型模型;低吻合度有V型胶原重塑模型、慢性移植物抗宿主病模型和Tsk-1/2鼠模型。结论:现有模型不能完全反应硬皮病疾病全过程状态,且其多是病理性模型,缺乏对中医证候的体现。故建立与临床吻合度较高的硬皮病病证结合动物模型及其评价指标有待进一步挖掘和完善,望对硬皮病临床及实验研究提供参考。
Objective:Based on the clinical characteristics of scleroderma in traditional Chinese and Western medicine,to analyze the existing animal models of scleroderma and provide ideas for the research of animal models of scleroderma.Methods:By consulting the relevant literature and reports on animal models with scleroderma at home and abroad,the clinical manifestations and diagnostic criteria of scleroderma in traditional Chinese and Western medicine were summarized.According to the clinical diagnostic indicators of scleroderma in traditional Chinese and Western medicine,the corresponding evaluation criteria were formulated.Each of the animal models and clinical core indicators of Western Medicine accorded with an assignment of 10%,and each of the relevant indicators accorded with an assignment of 4%.Each main symptoms accorded with an assignment of 15%,while each minor symptoms accorded with an assignment of 10%.To evaluate and analyze the clinical symptoms coincidence of existing animal models with scleroderma,and preliminarily put forward the evaluation index of animal models with scleroderma combined with TCM disease and syndrome.Results:It was found that the existing scleroderma animal models with high clinical coincidence were bleomycin-induced model and UCD200 chicken model,ROS-induced model and transgenic model were common coincidence models,V-type collagen remodeling model,chronic graft-versus-host disease model and Tsk-1/2 mouse model were low coincidence models.Conclusion:Existing models cannot fully reflect the whole process of scleroderma disease,and most of them are pathological models,lacking the manifestation of TCM symptoms.Therefore,it is necessary to further explore and improve the animal model with scleroderma combined with syndrome and its evaluation index in order to provide reference for clinical and experimental research of scleroderma.
