超声-微波辅助低共熔溶剂提取桑葚花青素及其活性研究

摘要 为优化超声-微波辅助低共熔溶剂提取桑葚中花青素的工艺条件,本试验以花青素的提取量为评价指标,在单因素试验的基础上进行正交试验,探讨低共熔溶剂类型、DESs含水量、料液比、超声温度、超声功率、超声时间、微波功率、微波时间对桑葚提取花青素的影响,并对其抗氧化活性进行研究。结果表明:花青素最佳提取条件为DESs含水量50%、料液比1:15 g/mL、超声温度60℃、超声功率300 W、超声时间30 min、微波功率750 W、微波时间40 s,在此条件下桑葚花青素的提取量可达8.892 mg/g。抗氧化活性试验表明,对DPPH自由基及羟基自由基的清除能力,桑葚花青素强于抗坏血酸(VC)。本研究可为低共熔溶剂的应用、桑葚花青素的提取及其相关产品的开发提供参考依据。 To optimize the process conditions for ultrasonic-microwave assisted eutectic solvent extraction of anthocyanins from mulberry,this experiment used anthocyanin extraction as the evaluation index,orthogonal test was carried out on the basis of single factor test,exploring the effect of the type of low eutectic solvent,DESs water content,solid-liquid ratio,ultrasonic temperature,ultrasonic power,ultrasonic time,microwave power and microwave time and studying antioxidant activity of anthocyanins from mulberry.The results showed that the optimal extraction conditions for anthocyanins were as follows:DESs water content 50%,solid-liquid ratio 1:15 g/mL,ultrasonic temperature 60℃,ultrasonic power 300 W,ultrasonic time 30 min,microwave power 750 W,microwave time 40 s.Under these conditions,the extraction amount of anthocyanins from mulberry could reach 8.892 mg/g.Antioxidant activity tests showed that the scavenging ability of mulberry anthocyanin to DPPH free radical and hydroxyl free radical was stronger than that of vitamin C.This study could provide reference for application of low eutectic solvent,extraction of mulberry anthocyanins and development of related products.
