高效液相色谱法测定人参属植物提取物中9种人参皂苷

Determination of 9 Ginsenosides in extracts of Panax species by high performance liquid chromatography

摘要 建立高效液相色谱法同时测定人参属植物中9种人参皂苷含量。经AgilentZORBAXSB-C18色谱柱分离,以乙腈-0.1%(体积分数)磷酸水溶液作为流动性进行梯度洗脱,流量为1.0mL/min,柱温为35℃,检测波长为203nm。结果表明,9种人参皂苷标准品在各自质量浓度范围内与色谱峰面积线性关系良好,相关系数均大于0.999,检出限为0.005~0.035mg/mL。在低、中、高三个浓度加标水平下,样品平均加标回收率为98.12%~101.94%,测定结果的相对标准偏差为2.50%~8.85%(n=6)。该方法简便快速、分离度好、重现性好、精密度高、结果可靠,可用于人参属植物中9种人参皂苷含量的同时测定。 A high performance liquid chromatography method was established for the simultaneous determination of 9 ginsenosides in Panax plants.The separation was performed on an Agilent ZORBAX SB-C18 column with gradient elution using acetonitrile-0.1%(volume fraction)phosphoric acid aqueous solution as the mobile phase.The flow rate was 1.0 ml/min,the column temperature was 35°C,and the detection wavelength was 203 nm.The results showed that the 9 Ginsenoside standards had a good linear relationship with the chromatographic peak area in their respective mass concentration ranges,the correlation coefficients were all more than 0.999,and the detection limits were 0.005-0.035 mg/mL.Under the three spiked levels of low,medium and high concentrations,the average spiked recoveries of the samples were 98.12%-101.94%,and the relative standard deviations of the determination results were 2.50%-8.85%(n=6).The method is simple and rapid,with good resolution,good reproducibility,high precision and reliable results.It can be used for the simultaneous determination of 9 Ginsenosides in Panax plants.
